I have a website and I want to optimize and insert keywords on the article. I want to know if it has a negative effect on the page itself? No it doesn't affect of anything as long as you don't stuff your keywords and you only limit the use of anchor text into 3 link. Make sure that you have follow the guidelines so that there will be no problem eventually. Actually even if you have submitted your article into a submission site you can even submit its link to a social bookmarking site after it has been approve to the article site you have been submitted. There is no one terms and condition by Google that if we can use meta tags after optimization it will effect to ranking. You can revise the content of a web page to improve the keyword density after published. Be sure that the keywords are applicable to the content of the page and not just spam to get higher in the results. One should always be reviewing their results and if they find that after a few months if what they were doing didn't work out, then make some adjustsment. But don't be impatient! Changing things every 5 mins does more harm than good.
Yes, Google formally announced that they are not using the meta keywords but that doesn't mean the "other guys" don't. e.g. Yahoo!'s How to Improve the Position of Your Website in Yahoo! Search Results specifically says:
