One of my sites at the moment is just a blog, so the blog is located at domainname.com.
I want to build a proper home page for the site, then have the blog at domainname.com/blog
Would this affect my ranking?
Thanks. Since pages are ranked and not sites, as such..........yes
Could go up.....more content....more traffic.......more links, etc.... If you use 301 permanent redirects for the blog articles the existing PR for the existing will eventually show.

Why don't you leave the blog where it is and make a home page through your blogging software? In Wordpress you can do this. Creating a Static Front Page - Codex @ Wordpress. That looks like it would be very basic and not give me much control over the design. I want to have a jquery featured content slider amongst other things, so looks like may have to take the hit on the PR in the short term to benefit long term. Yes ofcourse,it'll definitely affect your ranking,just remember whatever changes you 're doing in your site will be caught by google. Stretty,
